What It Does
PE-22-88 (sometimes referred to as PE-22-28) is derived from the NAP region of ADNP (Activity-Dependent Neuroprotective Protein) โ a transcription factor essential for brain development and neuroprotection. It enhances synaptic plasticity, promotes BDNF expression, and has demonstrated antidepressant-like and anxiolytic effects in preclinical models. Related to the broader NAP/ADNP peptide family, which also includes NAP (davunetide).
Mechanism of Action
- Microtubule stabilization: Interacts with microtubule end-binding proteins (EB1/EB3) โ stabilizes microtubule dynamics critical for synaptic function and axonal transport
- BDNF upregulation: Promotes BDNF expression โ synaptic plasticity, neuronal survival, and neurogenesis
- TREK-1 channel modulation: Inhibits TREK-1 (TWIK-related K+ channel) โ antidepressant mechanism complementary to SSRI/SNRI pathways
- Neuroprotection against tau/Aฮฒ: Protects against tau pathology and amyloid-beta toxicity
- ADNP pathway activation: Mimics core neuroprotective actions of full-length ADNP protein
