What It Does
Humanin is a small peptide encoded within the mitochondrial genome (16S rRNA region), making it one of a class called mitochondrial-derived peptides (MDPs). It is cytoprotective across multiple tissues — brain, heart, vasculature, and metabolic organs. Humanin levels decline with age and are inversely correlated with Alzheimer's disease risk and metabolic dysfunction. It is considered a key longevity signaling molecule and a bridge between mitochondrial health and systemic aging.
Mechanism of Action
- Anti-apoptotic signaling: Activates JAK2/STAT3, PI3K/Akt, and MAPK pathways → inhibits Bax-mediated mitochondrial apoptosis
- Neuronal protection: Blocks amyloid-beta (Aβ) toxicity in neurons; reduces tau pathology markers
- Insulin sensitization: Activates FPRL1/FPR2 and gp130 receptors → improves insulin sensitivity and glucose metabolism
- Mitochondrial function: Supports mitochondrial membrane potential and reduces ROS production
- IGF-1 modulation: Interacts with IGFBP-3, modulating IGF-1 bioavailability and longevity signaling
- Systemic cytoprotection: Cardioprotective, hepatoprotective, and vasculoprotective effects reported

Contraindications & Safety
- Active malignancy — anti-apoptotic signaling; theoretical concern
- Limited human data — extrapolated from rodent studies; use cautiously
- Drug interactions — JAK/STAT pathway involvement; caution with immunosuppressants
- Common side effects: Not well characterized in humans; generally well-tolerated in animal models
- Long-term safety profile in humans not established
Storage
- Lyophilized: -20°C long-term; 2–8°C up to 3 months
- Reconstituted: 2–8°C for 1–2 weeks; protect from light
- Solvent: Sterile/bacteriostatic water
