Mechanism of Action

DSIP is an endogenous nonapeptide originally isolated from rabbit thalamus by Monnier and colleagues in 1974. It promotes the transition to NREM Stage 3 (delta wave/slow-wave) sleep โ€” the deepest and most physiologically restorative sleep phase โ€” through mechanisms that remain incompletely characterized after decades of research. DSIP enhances nocturnal growth hormone secretion by amplifying the GH pulse that naturally occurs during slow-wave sleep, likely through somatostatin reduction at the hypothalamic level. It modulates the hypothalamic-pituitary-adrenal (HPA) axis by reducing ACTH release from the anterior pituitary, subsequently lowering cortisol levels and facilitating both sleep onset and stress reduction.
