What It Does
Argireline is the most widely used "Botox-like" topical peptide. It works by competitively inhibiting the SNARE complex โ the same molecular machinery that Botox (botulinum toxin) disrupts โ thereby reducing the release of neurotransmitters at the neuromuscular junction of facial muscles. The result is a reduction in muscle contraction intensity beneath expression lines (crow's feet, forehead, glabellar lines) without paralysis. Effects are reversible, dose-dependent, and require continuous use. At 10%, clinical studies show ~27โ30% reduction in wrinkle depth.
Mechanism of Action
- SNARE complex interference: Argireline's sequence mimics the N-terminal domain of SNAP-25 (synaptosomal-associated protein 25) โ competitively inhibits the SNAP-25/VAMP/Syntaxin SNARE complex โ reduces vesicular acetylcholine release at neuromuscular junctions
- Reduced muscle contraction: Less acetylcholine release โ lower facial muscle contraction amplitude โ reduced dynamic wrinkle formation
- Neurotransmitter catecholamine reduction: Also reduces catecholamine release in skin โ secondary relaxation effects
- Collagen synthesis support: Secondary effect โ reduces mechanical stress on dermal ECM โ slower collagen degradation from repetitive movement
