| Sample | Review covering Phase 2/3 clinical trial data for RGN-259 (Tβ4 eye drops) |
| Peptide(s) Studied | TB-500 |
Key Findings
- Phase 3 clinical trials of Tβ4 (as RGN-259) for dry eye and neurotrophic keratopathy were ongoing at time of publication
- Tβ4 eye drops demonstrated anti-inflammatory, anti-apoptotic, and wound healing properties in corneal models
- Clinical development represents the most advanced human therapeutic application of Tβ4
- Translational journey from basic actin-binding biology to clinical ophthalmology application documented
Study Design
Narrative review tracing the development of Tβ4 from bench science through Phase 2/3 clinical trials in ophthalmology. Covers preclinical mechanism of action, animal model data, and clinical trial design for RGN-259.
Limitations
- Phase 3 results not yet published at time of this review
- Ocular (topical) application may not generalize to systemic (injectable) use as TB-500
- Single therapeutic indication (ophthalmology) limits generalizability
Clinical Relevance
This is the closest to human clinical validation available for Thymosin Beta-4 in any indication. The Phase 2/3 trials for RGN-259 represent the highest level of clinical evidence for this peptide. Although the application is ophthalmological (topical eye drops), the underlying biology is relevant to systemic TB-500 use. Critical reference for the Evidence Level Summary of any TB-500 profile.
