What It Does

PE-22-88 (sometimes referred to as PE-22-28) is derived from the NAP region of ADNP (Activity-Dependent Neuroprotective Protein) โ€” a transcription factor essential for brain development and neuroprotection. It enhances synaptic plasticity, promotes BDNF expression, and has demonstrated antidepressant-like and anxiolytic effects in preclinical models. Related to the broader NAP/ADNP peptide family, which also includes NAP (davunetide).

Mechanism of Action

  1. Microtubule stabilization: Interacts with microtubule end-binding proteins (EB1/EB3) โ†’ stabilizes microtubule dynamics critical for synaptic function and axonal transport
  2. BDNF upregulation: Promotes BDNF expression โ†’ synaptic plasticity, neuronal survival, and neurogenesis
  3. TREK-1 channel modulation: Inhibits TREK-1 (TWIK-related K+ channel) โ†’ antidepressant mechanism complementary to SSRI/SNRI pathways
  4. Neuroprotection against tau/Aฮฒ: Protects against tau pathology and amyloid-beta toxicity
  5. ADNP pathway activation: Mimics core neuroprotective actions of full-length ADNP protein

Dosing Protocols

Standard Protocol

  • Dose: 1โ€“10 mg/day
  • Route: Intranasal (preferred for CNS access) or SubQ
  • Frequency: Once daily
  • Duration: 4โ€“8 week cycles
  • Timing: Morning preferred (stimulatory)
  • Source: The Peptides Bible, research literature

Intranasal Protocol

  • Dilute in sterile saline to appropriate concentration
  • 2โ€“3 drops per nostril, or use nasal atomizer

Stacks Well With

  • Semax – synergistic BDNF/neurotrophic factor stack
  • BDNF (exogenous or endogenous upregulators) – direct complement
  • Selank – anxiolytic balance to PE-22-88 cognitive stimulation
  • Dihexa – HGF + ADNP pathway synergy for cognitive enhancement
  • NSI-189 – hippocampal neurogenesis stack

Use Cases

Contraindications & Safety

  • Limited human data โ€” primarily preclinical; human safety not established
  • Stimulatory effects โ€” anxiety or insomnia if overdosed or taken late
  • Drug interactions โ€” TREK-1 modulation; caution with other psychiatric medications
  • Common side effects: Nasal irritation (intranasal route), overstimulation at high doses

Storage

  • Lyophilized: -20ยฐC long-term; 2โ€“8ยฐC short-term
  • Reconstituted solution: 2โ€“8ยฐC for 1โ€“2 weeks; protect from light
  • Solvent: Sterile saline (for intranasal) or bacteriostatic water (SubQ)
